1) Are the patches cumulative? I don't know if I ever got the first one... does the current one include the changes released in the first?

2) Does the AI use stellar manipulation? If I start a game with scattered "segments" that are unconnected and at low-tech, will the AI develop the tech needed and expand?

You only need the latest patch.
And yes, the AI should research and build stellar manipulation.
